What TGF-beta 1 measures, and why the report can mislead
An older report may mark a result high above 2,382 pg/mL. The current version of the same referred test uses an upper limit of 22,062 pg/mL. Confirm the date, lab, sample, test method, range, and platelet removal before linking the number to inflammation or mold illness.
Biology
A protein involved in immune and tissue responses
TGF-beta 1 helps regulate immune responses, cell growth, repair, and tissue remodeling. Many cell types make it, so a blood value cannot point to one organ or disease.
Collection
Platelets can manufacture a high result
Platelets store TGF-beta 1 and may release it after the draw. A sample that is not made platelet-free promptly may measure handling as much as biology.
Range
The same referred assay changed its interval
The former 344 to 2,382 pg/mL interval became 0 to 22,062 pg/mL in November 2024. The dated laboratory report outranks an old internet target.
Use
This is a specialist or research-context test
It may be used in selected cytokine, fibrosis, oncology, immune, or research questions. It is not a routine standalone test for brain fog, mold exposure, or CIRS.
A serum value, platelet-free EDTA plasma value, CTAD-plasma value, and proteomic relative-abundance measurement may all be called TGF-beta 1. They can't share one cutoff or one trend line as if they were one test.
Save this test
Save the assay with the number
My Fog keeps the dated range and platelet-free collection details beside the value. This prevents an old cutoff or a different sample from looking like a true change.
Repeat only to answer a defined clinical question with the same method and correct collection, not to get a number you prefer.
What can change how this result is read?
The current Mayo listing gives one method-specific interval, with no separate age or sex bands. Still, children, pregnancy, older age, and different specimens aren't interchangeable. So the report and the reason for testing matter more.
Infants and children
The 119-person reference study covered adults aged 18 to 63, not children. A child first needs a check for common causes, growth, nutrition, sleep, infection, medicines, vision, hearing, and development. Ask a child-health specialist what this test would change.
Teenagers and adults
Keep infection, vaccination, surgery, injury, exercise, smoking, alcohol, medicines, supplements, platelet count, and the original clinical question beside the result. A nonspecific cytokine should not replace basic blood work, examination, or cause-specific testing.
Women, pregnancy, and postpartum
Pregnancy studies report changing circulating TGF-beta 1, but they use different serum or plasma methods and are not a reference interval for this assay. Tell the obstetric clinician and laboratory about pregnancy or postpartum timing. New high blood pressure, severe headache, vision change, upper-abdominal pain, breathlessness, swelling, confusion, or weakness needs direct pregnancy care, not interpretation through this cytokine.
Men
The adult CTAD-plasma assay study did not find a significant sex difference, while large serum studies using other methods have reported sex and lifestyle associations. Use the method's printed interval and personal context instead of inventing a male target.
Older adults
Kidney, liver, lung, heart, autoimmune, cancer, platelet, and medicine contexts become more common with age and can matter more than the cytokine label. Review symptoms, standard organ tests, platelet count, medicines, and the specialist's reason for ordering before attributing a value to mold or cognitive symptoms.
How to get a usable platelet-free plasma result
Before the appointment, photograph the order and ask which lab will run the test. Confirm that it is human TGF-beta 1 in platelet-free EDTA plasma. Serum, platelet-rich plasma, tissue stains, genetic TGFB1 tests, and other TGF-beta types are different tests.
Ask the collection site to read the lab instructions before drawing blood. Mayo's current referred method uses a lavender-top EDTA tube. It requires two rounds of spinning, transfer to a plastic vial, freezing at minus 20 degrees C or colder, and frozen transport. The first spin must happen within 30 minutes.
A general website isn't a reason to fast, stop exercise, change food, or stop a medicine. The current Mayo instructions do not list patient fasting or exercise preparation. Follow the order and the performing laboratory, then record recent infection, vaccination, surgery, injury, hard exercise, pregnancy, medicines, supplements, platelet count, and the reason for testing.
If this is a repeat, use the same performing laboratory, specimen, method, and collection protocol when possible. Bring the earlier report. Copying only the value into a portal message isn't enough.
Confirm what was ordered
Match the name, specimen, performing laboratory, method, unit, and report date. TGF-beta 1 plasma, serum, proteomic relative abundance, and TGFB1 genetic testing are different measurements.
Use the laboratory's collection kit
The draw site should follow the performing laboratory's written centrifuge, transfer, freezing, and transport steps. This is not a routine spin-and-send plasma sample.
Remove platelets promptly
For Mayo's current referred assay, centrifuge the EDTA tube within 30 minutes, then centrifuge the separated plasma again. Platelets contain TGF-beta 1 and can release it after the blood leaves the body.
Freeze and ship correctly
Transfer platelet-free plasma to the specified plastic vial, freeze it at minus 20 degrees C or below, and keep it frozen in transport. Gross hemolysis, gross lipemia, contamination, or poor stability can lead to rejection.
Keep the report interval
Save the value, pg/mL unit, interval, assay, specimen, and performing lab together. Use the printed interval, not 2,380 pg/mL from an older report or CIRS website.
Ask what decision it changes
A specialist should name the condition in question, the other evidence required, and what a high, normal, failed, or repeated result would change. If no decision changes, repeating the cytokine may add cost without clarity.
How to understand a TGF-beta 1 result
Start with the report date, performing laboratory, specimen, assay, unit, and printed interval. Then check whether the sample was made platelet-free correctly. Only after those checks should the clinician interpret the value beside the reason for testing.
Inside this assay's interval
Inside the current report interval
The result falls inside that laboratory's method-specific interval. It does not rule out mold-related respiratory disease, a damp building, Long COVID, fibrosis, immune illness, or another cause of symptoms.
Near a limit or changed interval
Near a limit or discordant with an older report
Check whether the assay, specimen, processing, unit, and interval changed. A repeat is useful only when the same method and a correctly prepared platelet-free specimen answer a named clinical question.
Outside this assay's interval
Outside the current report interval
First confirm sample quality and the correct range. Then read an out-of-range result with the reason for testing, symptoms, exam, standard tests, and specialist review. The number has many possible causes.
A high number can be an old cutoff or a platelet problem
A report compared with 2,382 pg/mL may look alarming even though the current version of the same referred assay uses 22,062 pg/mL. Platelet activation can also add thousands of pg/mL outside the body. Calling the result inflammation and repeating treatment won't fix either problem.
See research details
These figures answer different questions. Keep each one with its exact specimen, method, population, and study design.
Keep the report date and performing laboratory beside the value. The former 2,382 upper limit isn't a timeless biological target, and it doesn't belong on a report made under the newer interval.
This proves why method and specimen matter. It doesn't replace the interval on a different assay, set a range for children or pregnancy, or give a treatment target.
A collection artifact can be much larger than the clinical difference someone is trying to interpret. Verify platelet removal and handling before building a diagnosis around the result.
Those figures belong to a selected research cohort, a multi-protein model, and relative proteomic measurements. They do not validate a single commercial TGF-beta 1 ELISA as a brain-fog or Long COVID diagnostic test.
Use visible dampness, water damage, musty odor, inspection, and supported respiratory or allergy assessment for the building and health questions. This blood test can't name a building, mold species, dose, or safe return date.
What you can do after a TGF-beta 1 result
There is no safe food, supplement, or home protocol that can be chosen from one TGF-beta 1 value. Protect the sample from handling errors, check common medical explanations, and address suspected building dampness or mold through inspection, repair and cleanup, not cytokine numbers.
Audit the report before paying for a repeat
Get the complete report and save the collection date, performing lab, specimen, method, value, unit, interval, and comments. If someone used 2,380 pg/mL, ask whether that cutoff matches the assay and date on your report.
Check the sample, not only the score
Ask whether EDTA plasma was made platelet-free with the required second spin, frozen promptly, and transported frozen. If those facts are unknown, the clinician and laboratory should decide whether the result is usable before anyone labels it inflammation.
Put common brain-fog checks ahead of a specialty cytokine
Bring the symptom timeline, sleep, medicines, alcohol and drug use, diet, menstrual or pregnancy context, recent infection, mood, pain, and neurologic changes. Ask whether CBC, metabolic, thyroid, iron, B12, glucose, sleep, medication, vision, or neurologic assessment fits the actual symptoms better.
Handle dampness at the source
If you see or smell dampness or mold, stop the moisture source, dry wet materials, and record the damage. Use a qualified inspector when the source is hidden or the job is large. A building check provides evidence that the blood result cannot.
This number alone isn't grounds to start or stop losartan, steroids, immune medicines, antifungals, binders, supplements, IV treatment, restrictive diets, or major mold cleanup. Chest pain, severe breathlessness, coughing blood, fainting, sudden confusion, facial droop, speech trouble, one-sided weakness, or a new severe headache needs urgent medical assessment.
What to save for a specialist visit
Keep these together
- Full dated report, performing laboratory, assay, specimen, value, pg/mL unit, and printed interval
- Tube, draw time, time to first spin, second spin, platelet-free status, transfer vial, freeze temperature, transport, and comments
- Platelet count, recent infection, vaccination, surgery, injury, exercise, pregnancy or postpartum timing, medicines, and supplements
- Reason for testing, symptoms and timing, building or exposure evidence, standard test results, and clinician interpretation
- What decision the test changed, whether a repeat is planned, and the exact method and conditions required for comparison
Question for the visit
“Was this the intended platelet-free assay, does the dated report use the current interval, could collection or platelets explain the value, what non-mold explanations matter, and what decision would a repeat change?”
